A brightfield compound microscope is a high power magnification optical instrument. Applications for this type of microscope are various, and include medical and clinical diagnostic laboratories for viewing blood, parasites, bacteria, pathogens, and microbes.
These microscopes are also used in education for study of microbiology and molecular biology. The inexpensive units are monocular, meaning it has only one eyepiece, and are used in elementary schools and in homeschool education. The cheap microscopes are still functional, but lack various components that are used to increase the quality of the optical image. For example, a cheap compound microscope probably will lack having a brightfield condenser, and may only have an aperature diaphragm with different size selectable openings for the light to pass.
A higher grade scope will have a full condenser on a gear system allowing it to move up or down as needed to focus the beam of light on the bottom of the slide containing the biological specimen. Higher end brightfield compound microscopes have light collectors in the base allowing optimal collection of the light and transmission to the brightfield condenser. They may also have focusing field diaphragms above the light collector. These features allow the light to be properly focused and controlled and to achieve Koehler illumination which is important for optimal imaging quality. The brightfield compound microscope uses multiple objectives on a rotating turret to change the magnification. The light source is located in the base of the microscope, and is typically a halogen bulb.
A brightfield light condenser is located under the stage and serves to condense the light and focus it on the bottom of the glass slide containing the specimen. Brightfield compound light microscopes are used to view prepared microscope slides. The thin slice or smear of the specimen is placed on the blank microscope slide, then a thin cover slip goes on top. Common objectives are 4x, 10x, 20x, 60x, and 100x. Each of these magnifications is multiplied by the eyepiece magnification to yield the final viewing optical magnification.
For example, with the above objectives, and with a 10x eyepiece pair, it would be 40x, 100x, 200x, 600x, and 1000x. For the 100x objective, it will generally be an oil immersion, requiring a tiny drop of oil to be placed on the microscope slide and the oil immersion objective to be brought into contact with the drop. This improves numerical aperature of the objectives and allows for higher resolution and resolving power to be obtained. Brightfield microscopes come as monocular, binocular, and trinocular units. The binocular models have a beam splitter prism that cuts the image equally and takes it to both oculars. This type of microscope is much easier to view than a simple cheap monocular microscope. The trinocular brightfield compound microscope has the added feature of a microphotography port in the top part of the head. This allows connectivity of still image cameras or full motion video cameras.
